چکیده

Embryonic hematopoiesis starts via the generation of primitive red blood cells (RBCs) that satisfy the embryo’s immediate oxygen needs. Although primitive RBCs were thought to retain their nuclei, recent studies have shown that primitive RBCs in mice enucleate in the fetal liver. It has been unknown whether human primitive RBCs enucleate, and what hematopoietic site might support this process. Our data indicate that the terminal maturation and enucleation of human primitive RBCs occurs in first trimester placental villi. Extravascular -globin primitive erythroid cells were found in placental villi between 5-7 weeks of development, at which time the frequency of enucleated RBCs was higher in the villous stroma than in circulation. RBC enucleation was further evidenced by the presence of primitive reticulocytes and pyrenocytes (ejected RBC nuclei) in the placenta. Extravascular RBCs were found to associate with placental macrophages, which contained ingested nuclei. Clonogenic macrophage progenitors of fetal origin were present in the chorionic plate of the placenta before the onset of fetoplacental circulation, after which macrophages had migrated to the villi. These findings indicate that placental macrophages may assist the enucleation process of primitive RBCs in placental villi, implying an unexpectedly broad role for the placenta in embryonic hematopoiesis. Erythropoiesis in the absence of adult hemoglobin.

During erythropoiesis, hemoglobin (Hb) synthesis increases from early progenitors to mature enucleated erythrocytes. Although Hb is one of the most extensively studied proteins, the role of Hb in erythroid lineage commitment, differentiation, and maturation remains unclear. In this study, we generate mouse embryos and embryonic stem (ES) cells with all of the adult α and β globin genes deleted ...

RED CELLS, IRON, AND ERYTHROPOIESIS miR-451 regulates zebrafish erythroid maturation in vivo via its target gata2

We demonstrate that in zebrafish, the microRNA miR-451 plays a crucial role in promoting erythroid maturation, in part via its target transcript gata2. Zebrafish miR-144 and miR-451 are processed from a single precursor transcript selectively expressed in erythrocytes. In contrast to other hematopoietic mutants, the zebrafish mutant meunier (mnr) showed intact erythroid specification but dimini...

RED CELLS, IRON, AND ERYTHROPOIESIS SCF induces -globin gene expression by regulating downstream transcription factor COUP-TFII

Increased fetal hemoglobin expression in adulthood is associated with acute stress erythropoiesis. However, the mechanisms underlying -globin induction during the rapid expansion of adult erythroid progenitor cells have not been fully elucidated. Here, we examined COUP-TFII as a potential repressor of -globin gene after stem cell factor (SCF) stimulation in cultured human adult erythroid progen...

Increased formation of early-labeled bilirubin in rats with iron deficiency anemia: evidence for ineffective erythropoiesis.

I RON DEFICIENCY ANEMIA is usually characterized by erythroid hyperplasia of the bone marrow, but impaired net synthesis of hemoglobin. This association suggests that the anemia may be due in part to ineffective erythropoiesis, i.e., destruction of immature red cells in the bone marrow or soon after their release into the circulation. Morphologic observations are also compatible with ineffectiv...

RED CELLS Remodeling the regulation of iron metabolism during erythroid differentiation to ensure efficient heme biosynthesis

Terminal erythropoiesis is accompanied by extreme demand for iron to ensure proper hemoglobinization. Thus, erythroblasts must modify the “standard” posttranscriptional feedback regulation, balancing expression of ferritin (Fer; iron storage) versus transferrin receptor (TfR1; iron uptake) via specific mRNA binding of iron regulatory proteins (IRPs). Although erythroid differentiation involves ...
